چکیده
347 Abstract: In this study the effect of diphtheria toxin (DT) on nitric oxide (NO) production from RAW 264.7 murine macrophages was investigated. Griess reagent was used to determine NO production by measuring nitrite levels in the culture supernatants. Corynebacterium diphtheriae G12/6 strain-produced DT (Limes flocculation activity and immunodiffusion assays were positive) demonstrated a dosedependent effect on RAW 264.7 macrophages to induce NO production. LNAME, an L-arginine analogue, inhibited NO production from DT-stimulated RAW 264.7 macrophages. At the given concentrations of DT, lipopolysaccharide (LPS or endotoxin)induced NO production from RAW 264.7 macrophages was also inhibited. RAW 264.7 macrophage cells, incubated with various concentrations of DT for 3 days, were killed by DT in a dose dependent manner. Endotoxin contamination of DT was demonstrated with limulus amoebocyte lysate assay. Polymyxin B, frequently added to neutralize the effects of LPS in vitro, inhibited DT-induced NO production from RAW 264.7 cells. The misleading data concerning NO inducing capacity of DT was seemed to be related to endotoxin contamination. Thus, DT does not seem to be capable of inducing NO production from macrophages.

Synergistic Effect of LPS, IFN- and Iron on Apoptosis of Balb/c Mice Macrophages Following Nitric Oxide Production
Objective(s) Previous studies have demonstrated that the nitric oxide (NO) dependent death of murine peritoneal macrophages activated in vitro with IFN-g and LPS is mediated through apoptosis. In the present study, we investigated the synergistic effect of LPS, IFN-g and iron on NO production and apoptosis. Materials and Methods After determination of iron cytotoxicity, the peritoneal macrop...
متن کاملEffect of Neutrophils on Nitric Oxide Production from Stimulated Macrophages
Background: During the initial phase of an infection, there is an upregulation of inducible nitric oxide synthase in the macrophages for the production of nitric oxide. This is followed by the recruitment of polymorphonuclear leukocytes (neutrophils) which release arginase. Arginase competes with inducible nitric oxide synthase for a common substrate L-arginine. Objective: To investigate whethe...
متن کاملInduction of inducible nitric oxide synthase increases the production of reactive oxygen species in RAW264.7 macrophages.
Macrophages produce a large volume of ROS (reactive oxygen species) through respiratory burst. However, the influence of iNOS [inducible NOS (nitric oxide synthase)] activation on ROS production remains unclear. In the present study, the kinetic generation of ROS in RAW264.7 murine macrophages was monitored by chemiluminescence. PMA induces a robust chemiluminescence in RAW264.7 cells, suggesti...
متن کاملThe anti-inflammatory effect of tussilagone, from Tussilago farfara, is mediated by the induction of heme oxygenase-1 in murine macrophages.
Tussilagone (TSL), isolated from the flower of buds of Tussilago farfara (Compositae), is a sesquiterpenoid that is known to exert a variety of pharmacological activities. In the present study, we demonstrated that TSL exerts anti-inflammatory activities in murine macrophages by inducing heme oxygenase-1 (HO-1) expression. Treatment of RAW264.7 cells with TSL-induced HO-1 protein expression in ...

Objective(s) The fractions of Candida albicans have been used as an immunomodulator. The present work assessed the effect of different fractions of C. albicans on nitric oxide (NO) production by mice peritoneal macrophages. Materials and Methods Cell wall and cytoplasmic fractions of C. albicans ATCC 10321 strain were extracted. Mice peritoneal macrophages were purified and cultured. Differen...
